The single player does run at 30FPS which some people will look down on, but I didn’t find it to hamper the experience. The animations and mo-capping are second-to-none. It really makes a difference and takes immersion to the next level. Whether it’s the way that puddles look when the light shines on them or the way that the shrubbery moves as you brush past it. Naughty Dog have gone out of their way to make absolutely everything in the environment look important. You’ll climb to new sites and have incredible views to take in. In terms of draw distance, Uncharted 4 takes it to a whole new level. The vast and varied landscapes are absolutely incredible and I often found myself stopping to take in the sights. Uncharted 4 is without a doubt the best looking game on PlayStation 4. It goes without saying that Naughty Dog are on top of the game when it comes to presentation. Uncharted 4: A Thief’s End really is a fitting end to Nathan Drake’s story. There are so many surprises and moments that pull at the heartstrings and I want to make sure that you experience that first hand. There are so many special moments that i’d like to pinpoint, but I really don’t want to spoil anything. They are integral to the story as a whole, and are believable in how they relate to the backstory of both Nathan and Sam Drake. Without a doubt, the villains in Uncharted 4 are the most memorable and well-written to date. Without going into too much detail, The Uncharted franchise has always struggled with creating memorable villains. It hit all the right notes and whilst it’s not the fast-paced non-stop story that we’ve come to know, it really does a great job of fleshing out the inner-workings of Nathan Drake and providing an insight into why he craves adventure and solving mysteries. I absolutely loved the way that the story was told. It focuses on Nathan Drake’s upbringing and the hardships that he has gone through in his earlier years. The narrative of Uncharted 4 is much more slow-paced than previous iterations. ![]() ![]() It doesn’t take long before things feel familiar and the Drake brothers are on the treasure hunt of their life. Nathan Drake is now settled into a semi-normal life with Elena when Nathan’s brother, Sam Drake returns from the dead. In our previous work, we considered the spatial variation in the gut microbiota in response to dietary CP manipulation and enterotoxigenic Escherichia coli exposure but have yet to explore temporal changes in ileal microbiota composition. The manipulation of dietary CP levels has been considered as a control measure for post-weaning diarrhoea, as its reduction has been shown to lower disease severity. In previous work, we have shown that both enterotoxigenic Escherichia coli (an important causative agent of post-weaning diarrhoea) exposure and an increase in dietary crude protein (CP) level have a significant impact on ileal microbiota composition in weaner pigs, in the absence of changes at faecal level. The immediate post-weaning period in pigs is a critical development stage, where disease risk is increased due to sudden changes in diet, environment and sow removal. However, there are few studies that explore temporal development and nutritional influences on the small intestinal microbiota. From previous work, we know that the porcine microbiota composition varies between gut sections and the impact of treatment effects have been investigated specifically on the ileal microbiota composition at single time points. Comparatively less studies have been done on the small intestinal microbiota, primarily due to the challenge of obtaining samples and the assumption that faecal material can be considered a proxy for upstream gut microbiota interactions. There have been many studies in recent years on the development of the porcine gut microbiota, which have primarily focussed on faecal microbial communities. These findings contribute to our understanding of a lesser studied gut compartment that is not only a key site of digestion, but also a target for the development of nutritional interventions to improve gut health and host growth performance during the critical weaning transition period. Nutritional composition influenced ileal microbiota development, with the high protein diet being associated with an increased abundance of significant porcine pathogens and the upregulation of GH33 exosialidases-which can influence host-microbe interactions and pathogenicity. The weaner pig ileal microbiota changed rapidly and was initially destabilised by the sudden introduction to feed. Samples from the low protein and high protein groups did not cluster separately by their CAZyme (carbohydrate-active enzyme) composition, but GH33 exosialidases were found to be significantly more abundant in the HP group (P = 0.006). The ileal microbiota was less stable in pigs fed the high protein diet (P = 0.05), with several pathogenic bacterial genera being significantly higher in abundance in this group. The ileal microbiota became less stable after the introduction to a solid diet at weaning (P = 0.036), was highly variable between pigs and no relationship was observed between average daily weight gain and microbiota composition. We identified significant changes in microbiome structure (P = 0.01), a reduction in microbiome richness (P = 0.02) and changes in the abundance of specific bacterial taxa from baseline until 13 days post-weaning. The remaining pigs were introduced to either an 18% (low) or 23% (high) protein diet on weaning day (day 0) and further ileal digesta sampling was carried out at days 5, 9 and 13 post-weaning. Ileal digesta samples were obtained at post-mortem prior to weaning day (day − 1) for baseline measurements. Here, the development of the porcine ileal microbiota under differing levels of dietary protein was observed over the immediate post-weaning period.
